The story begins with Lightning McQueen (voiced by Owen Wilson) returning to Radiator Springs after winning his fourth Piston Cup. He reunites with his best friend, Mater (voiced by Larry the Cable Guy), the goofy and kind-hearted tow truck. Lightning is soon invited to participate in the World Grand Prix, a prestigious international race sponsored by Miles Axlerod (voiced by Eddie Izzard), an oil tycoon who has developed a new environmentally friendly fuel called Allinol.
The World Grand Prix consists of three races in different locations Tokyo, Japan Porto Corsa, Italy and London, England. Lightning, with Mater as his crew chief, faces stiff competition from racers like the cocky Italian Formula One car Francesco Bernoulli (voiced by John Turturro).
Meanwhile, the plot thickens as Mater inadvertently gets caught up in a spy mission. British spy Finn McMissile (voiced by Michael Caine), a suave Aston Martin, and his rookie partner Holly Shiftwell (voiced by Emily Mortimer) mistake Mater for an American secret agent. They are investigating a conspiracy involving a group of lemon cars (older, less reliable models) who are planning to sabotage the World Grand Prix.
As the races unfold, Mater, Lightning, and the spies uncover a plot to use Allinol to cause the race cars to explode, discrediting alternative fuels and increasing demand for gasoline. The mastermind behind the plot is revealed to be Professor Zündapp (voiced by Thomas Kretschmann), a scientist working for the lemon cars, and Sir Miles Axlerod himself, who secretly profits from the oil industry.
In the end, Mater, with help from Finn and Holly, manages to thwart the villains plan. Lightning McQueen wins the final race in London, but the real victory comes when Mater is recognized as a hero for his role in stopping the conspiracy. Mater is knighted by the Queen of England, and the film concludes with the residents of Radiator Springs celebrating their victory and the return of their friends.
